Type I Importers

Type I importers are ATP-binding cassette (ABC) membrane transport systems that move specific nutrients and ions into cells, supporting growth when substrates are scarce. A soluble substrate-binding protein first captures the cargo and delivers it to transmembrane domains, where ATP binding and hydrolysis drive conformational changes that open the transporter alternately to the outside and cytoplasm. In bacteria, Type I importers commonly mediate uptake of sugars, amino acids, peptides, and metal ions. Their substrate selectivity, energy-coupling mechanism, and role in nutrient acquisition make them important for understanding microbial physiology, host interactions, antimicrobial targets, and engineered transport systems.

Here we describe a new method to study protein import into isolated chloroplasts under stress. The method is rapid and straightforward, and can be applied to study the consequences of different stress conditions for chloroplast protein import, and the corresponding regulatory mechanisms.
