Valve Leaflet Dissection

Valve leaflet dissection is a biological and anatomical technique used to isolate individual heart-valve leaflets for detailed structural and functional study. Researchers carefully separate the leaflet from surrounding valve tissue and supporting structures, preserving its layered architecture for examination of extracellular matrix, cellular organization, and mechanical properties. The method supports investigations of valve development, tissue remodeling, disease-related changes, and biomechanical behavior. Findings from dissected leaflets can help clarify how valves regulate blood flow, identify structural features associated with dysfunction, and inform research on cardiovascular disease, biomaterials, and tissue-engineered heart-valve replacements.

2013

There has been renewed interest in developing polymer valves. Here, the objectives are to demonstrate the feasibility of modifying a commercial pulse duplicator to accommodate tri-leaflet geometries and to define a protocol to present polymer valve hydrodynamic data in comparison to native and prosthetic valve data collected under near-identical conditions.

This protocol describes the isolation, culture, and calcification of rat-derived valve interstitial cells, a highly physiological in vitro model of calcific aortic valve disease (CAVD). Exploitation of this rat model facilitates CAVD research in exploring the cell and molecular mechanisms that underlie this complex pathological process.

2022

The presented method offers an innovative way for engineering biomimetic fiber structures in three-dimensional (3D) scaffolds (e.g., heart valve leaflets). 3D-printed, conductive geometries were used to determine shape and dimensions. Fiber orientation and characteristics were individually adjustable for each layer. Multiple samples could be manufactured in one setup.
