Vasculogenesis

Vasculogenesis is the de novo formation of blood vessels from mesoderm-derived precursor cells, establishing the first vascular networks during embryonic development and supporting tissue growth. During this process, angioblasts differentiate into endothelial cells, aggregate into cords, and organize into interconnected vascular channels that later undergo remodeling and maturation. In biology, vasculogenesis provides a framework for understanding embryonic patterning, organ development, and the origins of the circulatory system. Studying this process also informs research on tissue engineering, regenerative medicine, cancer biology, and vascular disorders, where controlling new vessel formation may improve tissue integration or reveal disease mechanisms.

Here, we present a protocol for time-lapse imaging and analysis of vasculogenesis in vitro using phase contrast microscopy coupled with the open source software, Kinetic Analysis of Vasculogenesis. This protocol can be applied to quantitatively assess the vasculogenic potential of numerous cell types or experimental conditions that model vascular disease.
