Viral Rescue

Viral rescue is the laboratory recovery of infectious virus from viral genetic material, such as a cloned DNA or complementary DNA genome, and is a key method in modern virology. Researchers introduce the genome, often with helper plasmids encoding essential viral proteins, into permissive cells, where cellular and viral enzymes drive genome replication, protein production, and assembly of new virus particles. The rescued virus can then be characterized to examine gene function, replication, host range, and pathogenic mechanisms. Viral rescue also supports the development of vaccine candidates, viral vectors, and antiviral strategies, while enabling controlled study of engineered mutations under appropriate biosafety conditions.

Capsulorhexis is an important step in phacoemulsification surgery. A surgeon creates a continous curvilinear tear on the anterior lens capsule by controlling the tearing vector forces. A peripherally extended tear is a serious complication. This video demonstrates a novel technique of rescuing capsular radial tear out using a cystotome.

(2015)This video demonstrates the recovery of infectious Japanese encephalitis virus from synthetic RNA transcribed from a bacterial artificial chromosome clone. It shows how cells are electroporated with the synthetic RNA, allowing the viral genome to direct the production of new virions. The resulting plaques formed in infected cell monolayers confirm...

Viral Recombination

2019

Cells are sometimes infected by more than one virus at once. When two viruses disassemble to expose their genomes for replication in the same cell, similar regions of their genomes can pair together and exchange sequences in a process called recombination. Alternatively, viruses with segmented genomes can swap segments in a process called reassortment. Viral Recombination Can Create New Diseases Some diseases can infect multiple species. For example, pigs can be infected by some human and...
