Wash Step Protocol

A Wash Step Protocol is a standardized laboratory procedure for rinsing biological samples, cells, or assay surfaces to remove unwanted material while preserving the target of interest. It typically introduces a suitable wash buffer, allows brief contact with the sample, and removes the liquid through aspiration, decanting, or centrifugation; repeated cycles use dilution and physical separation to clear unbound reagents, debris, or contaminants. In biology, careful washing supports immunoassays, cell staining, nucleic acid workflows, and other protocols by lowering nonspecific background and improving signal reliability. Buffer composition, temperature, duration, and handling force must match the sample and assay.

2016

Retinal pigment epithelium (RPE) replacement strategies and gene-based therapy are considered for several retinal degenerative conditions. For clinical translation, large eye animal models are required to study surgical techniques applicable in patients. Here we present a rabbit model for subretinal surgery geared towards RPE transplantation, which is versatile and cost-efficient.

The adult Drosophila brain is a valuable system for studying neuronal circuitry, higher brain functions, and complex disorders. An efficient method to dissect whole brain tissue from the small fly head will facilitate brain-based studies. Here we describe a simple, one-step dissection protocol of adult brains with well-preserved morphology.
