Yolk Sac Isolation

Yolk sac isolation is a dissection technique that separates the embryonic yolk sac, an extraembryonic membrane involved in early nutrient exchange, blood formation, and vascular development, from the developing embryo. Under a stereomicroscope, researchers collect embryos at a defined developmental stage and use fine tools to remove surrounding membranes and detach the yolk sac while preserving its tissue layers and cellular organization. Isolated yolk sacs can then be cultured, stained, or analyzed for hematopoietic progenitors, blood vessel formation, gene expression, and responses to experimental treatments. This method supports studies of embryonic development, congenital abnormalities, cell differentiation, and developmental toxicity.

The lack of tools to measure material properties and tensional parameters in vivo prevents validating their roles during development. We employed atomic force microscopy (AFM) and nanoparticle-tracking to quantify mechanical features on the intact zebrafish embryo yolk cell during epiboly. These methods are reliable and widely applicable avoiding intrusive interventions.
