Agarose Injection Plate

An Agarose Injection Plate is a laboratory platform that immobilizes small biological specimens during precise microinjection, making it useful for experimental cancer research. The plate uses a thin layer or molded wells of hydrated agarose, a porous, semisolid polysaccharide, to hold specimens in position while permitting access by a fine injection needle without preventing gas or fluid exchange. In cancer studies, researchers can use agarose injection plates to introduce tumor cells or other experimental materials into model organisms, supporting the development of xenograft assays and measurements of tumor growth, invasion, metastasis, or treatment response. This preparation improves injection consistency and experimental reproducibility.

Source: Mohan, R., et al. Intramuscular Injections Along the Motor End Plates: A Minimally Invasive Approach to Shuttle Tracers Directly into Motor Neurons. J. Vis. Exp. (2015)This video demonstrates the targeted delivery of a fluorescent neuronal tracer into motor neurons via intramuscular injection at the motor end plates (MEPs). It outlines the steps involved in preparing the rat, injecting the tracer, and assessing labeled motor neurons for neurotherapeutic potential.

The efficacy of intramuscular uptake and retrograde transport of molecules to corresponding motor neurons depends on the location of the injection sites with respect to the motor end plates (MEPs). Here, we describe how to locate MEPs on skeletal muscles to optimise retrograde transport of tracers into motor neurons.

Source: Denaxa, M., el al., Transplantation of Chemogenetically Engineered Cortical Interneuron Progenitors into Early Postnatal Mouse Brains. J. Vis. Exp. (2019)This video demonstrates the procedure for injecting a plasmid vector carrying the gene of interest into interneuron progenitors in an embryonic brain slice, followed by electroporation for plasmid entry and gene expression. These modified progenitors can later be used for treating brain disorders.
