Scaffold Seeding

Scaffold seeding is a technique that introduces living cells onto or into a three-dimensional biomaterial scaffold, creating an organized environment for growth and study. Researchers apply a defined cell suspension to the scaffold, where cells attach to its surfaces or pores and progressively distribute through the structure under suitable culture conditions. In cancer research, seeding tumor or stromal cells onto scaffolds helps recreate aspects of the tumor microenvironment, including cell interactions and spatial organization. These models can support investigations of cancer growth, invasion, treatment response, and biomaterial-based approaches to tissue engineering.

MSC Seeded Scaffold Preparation: A Technique to Prepare Scaffold Seeded with Mesenchymal Stem Cells for Surgical Implantation into Murine Model

This video describes the technique of seeding mesenchymal stem cells (MSCs) onto a biodegradable poly(lactic acid) (PLA) scaffold. The prepared scaffolds can be implanted into the surgical resection cavity of the brain from where they may migrate towards tumor foci. Further, these cells can be engineered to express therapeutic proteins to stimulate apoptosis in cancer cells.

Engineered tissues heavily rely on proper vascular networks to provide vital nutrients and gases and remove metabolic waste. In this work, a stepwise seeding protocol of endothelial cells and support cells creates highly organized vascular networks in a high-throughput platform for studying developing vessel behavior in a controlled 3D environment.

Here, we present a protocol to fabricate electrospun nanofiber scaffolds with gradated organization of fibers and explore their applications in regulating cell morphology/orientation. Gradients with regard to physical and chemical properties of the nanofiber scaffolds offer a wide variety of applications in the biomedical field.
