Tumor Cell Tracking

Tumor cell tracking is the measurement and visualization of cancer cell location, movement, and fate over time, helping researchers understand how tumors grow and spread. In common approaches, cells are marked with fluorescent dyes, reporter proteins, or genetic barcodes and then monitored by microscopy or whole-animal imaging; changes in signal reveal migration, proliferation, survival, or treatment response. In cancer research, tracking distinguishes invasive cells from stationary populations, maps metastatic dissemination, and connects cellular behavior with tumor microenvironment conditions. These methods support studies of tumor progression, therapeutic resistance, and the development of strategies that target cancer cell movement and survival.

An In Vivo Technique to Track Bispecific Antibody-Induced T Cell Trafficking to Tumors

This video demonstrates a method for tracking bispecific antibody-induced T cell trafficking to tumors in a mouse model. Luciferase-transduced effector T cells are injected into a mouse with a subcutaneous tumor cell xenograft. Bispecific antibodies are then introduced, binding to T cells and the tumor cells. The localization of the T cells is tracked using bioluminescent imaging, with the magnitude of the signal indicating the extent of T cell infiltration into the tumor.

PTM Based Tumor Microenvironment Monitoring: A Method for Tracking Changes in ECM Rigidity Using Tumor Spheroids

This protocol describes a technique for monitoring extracellular matrix rigidity changes by using particle-tracking microrheology (PTM). This is done by culturing 3D pancreatic tumor spheroids into ECM with embedded fluorescent probes for video particle-tracking and analysis.

Stereotactic ablative body radiation therapy (SBRT) involves the precise delivery of high-dose radiation to cancer tumor targets. A novel SBRT platform offers a first-of-its-kind gimbaled radiation accelerator mounted within a pivoting O-ring gantry capable of dynamic image-guided tumor tracking. Here, we describe dynamic tumor tracking for lung targets.

We established the photoconvertible PSmOrange system as a powerful, straight-forward and cost inexpensive tool for in vivo cell tracking in GFP transgenic backgrounds. This protocol describes its application in the zebrafish model system.
