Tumor Prevention Models

Tumor prevention models are experimental systems used to study how cancers begin and to evaluate strategies that may stop or delay tumor formation. In cancer research, these models reproduce key stages of carcinogenesis by exposing cells, tissues, or organisms to cancer-promoting conditions and then measuring genetic changes, abnormal growth, and progression toward malignancy. Researchers use them to test preventive agents, investigate risk factors, and identify biological pathways involved in tumor initiation. Findings from cellular, organoid, and animal models can guide biomarker development and the design of prevention studies, while also revealing how effectively potential interventions work across different cancer types.

Evaluation of Tumor-infiltrating Leukocyte Subsets in a Subcutaneous Tumor Model

This protocol describes a method for the detailed evaluation of leukocyte subsets within the tumor microenvironment in a mouse tumor model. Chemerin-expressing B16 melanoma cells were implanted subcutaneously into syngeneic mice. Cells from the tumor microenvironment were then stained and analyzed by flow cytometry, allowing for detailed leukocyte subset analyses.

This video demonstrates a cell-based tumor vaccination assay. A mouse, intradermally injected with B16-F10 melanoma cells for tumor formation, is vaccinated with B16-F10 cells secreting Flt3L cytokine near the developed tumor. The secreted Flt3L from the vaccine promotes the maturation and proliferation of dendritic cells that then present tumor antigens to T cells in lymph nodes, triggering a targeted anti-tumor immune response.
