Auxiliary Complexing Agents

Auxiliary complexing agents are ligands that bind metal ions alongside a primary ligand, helping control the composition, stability, and behavior of coordination complexes in chemistry. They work through donor atoms that coordinate with a metal center, often competing with solvent molecules or other ligands and altering metal-ion speciation, solubility, reaction kinetics, and selectivity; their effects can depend strongly on pH and concentration. These agents support analytical titrations, metal-ion separations, extraction processes, coordination synthesis, and industrial treatment of metal-containing solutions. By tuning complex formation, they improve measurement accuracy and help researchers control challenging inorganic and biochemical reactions.

EDTA titrations are usually carried out in highly basic conditions, where the fully deprotonated form of EDTA, Y4−, actively complexes with the free metal ions in the solution. We demonstrate the metalation, purification, and characterization of lanthanide complexes. The complexes described here can be conjugated to macromolecules to enable tracking of these molecules using magnetic resonance imaging.

Auxiliary liver transplantation provides a temporary support in acute hepatic failure, until regeneration of the failing liver. The heterotopic auxiliary liver transplantation (HALT) with portal vein arterialization (PVA) renders sufficient liver function. We developed an analogous technique in the rat, to examine the influence of the portal vein arterialization on the morphology and function of the graft.

This protocol describes the preparation and characterization of a dendrimeric magnetic resonance imaging (MRI) contrast agent that carries cyclen-based macrocyclic chelates coordinating paramagnetic gadolinium ions. In a series of MRI experiments in vitro, this agent produced an amplified MRI signal when compared to the commercially available monomeric analogue.
