Chain Extension Method

The chain extension method is a synthetic strategy in chemistry that lengthens a molecular framework by adding one or more atoms, often carbon atoms, to an existing structure. It typically uses a reactive functional group, organometallic reagent, or coupling partner to form a new covalent bond, followed by functional-group conversion when needed to preserve or introduce the desired reactivity. In organic synthesis, chain extension supports the preparation of homologous compounds, complex natural products, pharmaceuticals, and advanced materials. Careful control of reagent compatibility and reaction conditions helps determine the length, connectivity, and functional groups of the resulting molecule.

This video demonstrates an assay for promoting the growth of neuron projections known as neurite outgrowth using the macrophage-conditioned medium. Co-culture neurons and macrophages with cAMP and harvest the macrophage-derived conditioned medium. Growing the neurons with the conditioned medium promotes neurite outgrowths. These outgrowths are fluorescently labeled and visualized using a fluorescence microscope.
