Dna Separation

DNA separation is the chemical and analytical process of resolving DNA molecules into distinct fractions based on properties such as size, charge, or binding behavior. In gel electrophoresis, an electric field drives the negatively charged phosphate backbone toward a positive electrode, while a porous agarose or polyacrylamide matrix slows larger fragments more than smaller ones, producing separated bands. These patterns allow researchers to estimate fragment length, assess sample quality, and isolate DNA for further analysis. DNA separation supports molecular biology, genetics, forensic testing, and biochemical research by making complex nucleic acid mixtures easier to identify, compare, and manipulate.

We describe an efficient method to separate single-stranded DNA, double-stranded DNA and RNA molecules from environmental viral communities. Nucleic acids are fractionated using hydroxyapatite chromatography with increasing concentrations of phosphate-containing buffers. This method permits the isolation of all viral nucleic acid types from environmental samples.

Dialysis: Diffusion Based Separation

2023

Dialysis is a common technique used in biochemistry for separating molecules based on diffusion. In this procedure, a semipermeable membrane allows the movement of certain molecules based on size. This method can be applied to the removal of buffer, known as desalting, or exchanging buffer molecules or ions from a protein solution. This video covers the principles of dialysis along with a general procedure. Several applications of dialysis are reviewed, including the removal of gradient...
