Headspace Vial

A headspace vial is a sealed container used to analyze volatile compounds in liquid or solid samples by sampling the gas above the specimen rather than injecting the matrix itself. In chemistry, heat and agitation can drive volatile molecules from the sample into the headspace, where they partition according to vapor pressure, temperature, and matrix interactions. This gas phase is then transferred to a gas chromatograph, commonly by an automated sampler, for separation and detection. Headspace vials reduce contamination from nonvolatile materials, simplify preparation, and support measurements in environmental analysis, pharmaceuticals, food chemistry, and forensic science, while controlled equilibration improves reproducibility.

The overall goal of this protocol is to measure greenhouse gas emissions from paddy fields using the static closed chamber technique. The measurement system needs specific adjustments due to the presence of both a permanent water layer in the field and of the plants within the chamber headspace.
