Irreversible Enolate

An irreversible enolate is an enolate ion formed when a strong base removes an α-hydrogen from a carbonyl compound under conditions that prevent effective reprotonation, making deprotonation effectively one-way. Typically, a non-nucleophilic base such as lithium diisopropylamide (LDA) deprotonates the less hindered α-position at low temperature, and the resulting lithium enolate is rapidly trapped by an electrophile. This kinetic control enables selective carbon–carbon bond formation through reactions such as alkylation, acylation, and aldol addition. In synthetic chemistry, irreversible enolate formation provides predictable regioselectivity and helps construct complex molecules from aldehydes, ketones, and related carbonyl compounds.
