Neutral Red Methylene Blue

Neutral red methylene blue is a vital staining system that combines two chemically responsive dyes to reveal differences in cell viability and intracellular conditions. Neutral red enters living cells and concentrates in acidic compartments such as lysosomes, producing a red signal, while methylene blue stains nonviable cells more strongly and can shift between blue and colorless forms as its oxidation state changes. In chemistry and biological analysis, this paired stain supports rapid microscopic assessment of cell survival, membrane integrity, and metabolic activity. Its contrasting colors help researchers evaluate cultures, compare experimental treatments, and monitor cellular responses without immediately destroying the sample.

In this video, we demonstrate in vitro staining of neutral lipids in hepatocytes to detect hepatic steatosis or excessive fat accumulation in liver. Excessive fat is accumulated in the cells in form of lipid droplets. Oil Red O is a lipid soluble dye that binds to intracellular neutral lipids in these lipid droplets. When observed under a light microscope, these intracellular lipid droplets appear intensely red in color.
