Pre-targeting Imaging

Pre-targeting imaging is a chemical imaging strategy that separates delivery of a targeting molecule from administration of an imaging probe, helping visualize specific biological sites while limiting background signal. In a typical approach, a targeting vector first binds a cellular marker and presents a reactive handle or binding site; a subsequently administered probe then recognizes that handle through high-affinity binding or a bioorthogonal reaction, while unbound probe clears from circulation. This two-step design can improve image contrast, reduce exposure of healthy tissues, and support the use of short-lived radioisotopes or fluorescent labels. In chemistry, it guides the development of selective reactions, linkers, and molecular imaging agents.

This video describes the technique of pretargeted radioimmunotherapy or PRIT, in which immunoconjugates followed by radioligands are administered for treating tumors. This radioimmunotherapy reduces radiation doses to healthy tissues and utilizes radionuclides with very low half-lives incompatible with antibody-based vectors.

The bioorthogonal inverse electron demand Diels-Alder cycloaddition has been harnessed to create an effective and modular pretargeted PET imaging strategy for cancer. In this protocol, the steps of this methodology are described in the context of a model system employing the colorectal cancer targeted antibody huA33 and a 64Cu-labeled radioligand.

This protocol describes the synthesis and characterization of a trans-cyclooctene (TCO)-modified antibody and a 177Lu-labeled tetrazine (Tz) radioligand for pretargeted radioimmunotherapy (PRIT). In addition, it details the use of these two constructs for in vivo biodistribution and longitudinal therapy studies in a murine model of colorectal cancer.

We describe a protocol for hybrid imaging, combining fluorescence-mediated tomography (FMT) with micro computed tomography (µCT). After fusion and reconstruction, we perform interactive organ segmentation to extract quantitative measurements of the fluorescence distribution.
