Razor Blade Cutoff

Razor blade cutoff is a mechanical sample-preparation approach that uses a sharp blade to create a controlled cut in a chemical sample, film, or coating. By concentrating force along a narrow edge, the blade promotes localized shear or fracture, producing a defined cross-section while limiting disturbance to nearby layers. In chemistry, this cutoff can support inspection of film thickness, layer structure, interfaces, and mechanical failure, particularly during characterization of polymers and surface coatings. Its value lies in linking a simple preparation step with reliable structural and materials analysis.

Preparation of Adult Drosophila Eyes for Thin Sectioning and Microscopic Analysis

A standard approach to prepare adult Drosophila eyes for semi-thin sectioning and light microscopic analysis is presented here. The protocol can be used for gross morphological analysis of eye defects, or with the indicated adjustments can be used to determine genetic requirements of genes in specific cell types of the eye (e.g. clonal analysis of photoreceptors) or for electron microscopic analysis.

Traditional procedures for the isolation of soluble type 1 collagen (COL1) require about 10 days from start to finish because of lengthy buffer incubations and laborious resuspensions of fibrils. Here, we describe a means to purify COL1 from small dermal biopsies in less than 3 hr.
