Sample Characterization

Sample characterization is the systematic measurement of a material’s composition, structure, physical properties, and chemical behavior to determine what it contains and how it functions. In chemistry, researchers combine techniques such as spectroscopy, chromatography, microscopy, elemental analysis, and thermal analysis, selecting methods that probe interactions between the sample and electromagnetic radiation, separation media, heat, or electrons. Comparing measurements with standards, reference spectra, and calibration data helps identify compounds, quantify components, and assess purity, morphology, stability, or phase. These results support reaction development, materials research, pharmaceutical quality control, environmental analysis, and reproducible interpretation of experimental findings.

In this video, we describe a protocol for the immunostaining and confocal microscopy-based visualization of macrophage extracellular traps (METs) in murine lung tissue. Tissue-resident macrophages are characterized by the expression of the macrophage-specific F4/80 surface glycoprotein marker, while METs are visualized by labeling the MET markers matrix metalloproteinase 9, citrullinated histone H3, and extracellular chromatin.

Isolation and Functional Characterization of Human Ventricular Cardiomyocytes from Fresh Surgical Samples

Current knowledge on the cellular basis of cardiac diseases mostly relies on studies on animal models. Here we describe and validate a novel method to obtain single viable cardiomyocytes from small surgical samples of human ventricular myocardium. Human ventricular myocytes can be used for electrophysiological studies and drug testing.
