Clearance Kinetics

Clearance kinetics describes how quickly the body removes a drug or other substance from the circulation over time, making it central to pharmacokinetics and clinical dose planning. It interprets concentration changes through processes such as hepatic metabolism and renal excretion: in first-order elimination, a constant fraction is removed per unit time, whereas capacity-limited pathways can produce zero-order behavior when metabolic or transport systems become saturated. Clinicians use clearance estimates with volume of distribution and half-life to select loading and maintenance doses, adjust therapy for renal or hepatic impairment, and anticipate drug accumulation or interactions, supporting safer, individualized treatment.

A Split-Luciferase Complementation Assay for Temporally Resolved Measurement of Tau Clearance in Microglia

2026

We describe a sensitive, time-resolved quantitative platform to measure clearance of extracellular tau in human induced pluripotent stem cell (iPSC)-derived microglia using a microplate-based luminescence system. This approach enables sensitive detection of tau degradation kinetics in a microwell format, allowing for high-throughput interrogation of tau handling under various experimental conditions.

Hepatic Drug Clearance: Restrictive and Nonrestrictive Clearance

2025

Hepatic clearance refers to the volume of blood cleared of a drug by the liver per unit of time. It plays a crucial role in drug metabolism and elimination. While hepatic clearance is commonly estimated by subtracting renal clearance from total body clearance, other pathways, such as pulmonary or biliary clearance, may also contribute. However, these pathways are generally less significant than hepatic and renal clearance. Most drugs undergo restrictive clearance, which is proportional to the...

Kinetic Molecular Theory: Molecular Velocities, Temperature, and Kinetic Energy

The kinetic molecular theory qualitatively explains the behaviors described by the various gas laws. The postulates of this theory may be applied in a more quantitative fashion to derive these individual laws. Collectively, the molecules in a sample of gas have average kinetic energy and average speed; but individually, they move at different speeds. Molecules frequently undergo elastic collisions in which the momentum is conserved. Since the colliding molecules are deflected off at different...

Enzyme Assays and Kinetics

2023

Enzyme kinetics describes the catalytic effects of enzymes, which are biomolecules that facilitate chemical reactions necessary for living organisms. Enzymes act on molecules, referred to as substrates, to form products. Enzyme kinetic parameters are determined via assays that directly or indirectly measure changes in substrate or product concentration over time. This video will cover the basic principles of enzyme kinetics (including rate equations) and kinetic models. The concepts governing...

Take pig kidney cell cultures, with one culture pre-treated with an inhibitor that increases lysosomal pH, impairing autophagic-lysosomal degradation.Add pathogenic bacteria and centrifuge to enhance host-pathogen contact.Incubate to allow bacterial invasion and internalization into endocytic vesicles. The internalized bacteria consume host nutrients to replicate, triggering metabolic stress. This stress activates autophagy, where an autophagic membrane expands around the vesicle, forming an...
