Hydrophobic Matrices

Hydrophobic matrices are solid materials designed to interact selectively with nonpolar regions of biomolecules, making them useful for separating and purifying proteins in clinical research and bioprocessing. In hydrophobic interaction chromatography, proteins bind to hydrophobic ligands on the matrix under high-salt conditions, which promote exposure and association of hydrophobic surfaces; reducing salt concentration or adding suitable modifiers then releases the bound molecules. These matrices support the purification of therapeutic proteins, antibodies, vaccines, and diagnostic reagents while helping preserve biological activity. Their selectivity, scalability, and compatibility with aqueous processing make them important tools in pharmaceutical manufacturing and clinical laboratory workflows.

In this video, we demonstrate the purification of calcium-binding protein from a dialyzed cell lysate through calcium-dependent hydrophobic interaction chromatography. The calcium-binding proteins expose a hydrophobic region upon binding with calcium, facilitating interaction with a hydrophobic group on resin. Later these proteins are eluted using calcium chelator EDTA that reverses the interaction.

The BoTest Matrix botulinum neurotoxin (BoNT) detection assays rapidly purify and quantify BoNT from a range of sample matrices. Here, we present a protocol for the detection and quantification of BoNT from both solid and liquid matrices and demonstrate the assay with BOTOX, tomatoes, and milk.
