In Situ Perfusion

In situ perfusion is a technique that circulates a controlled fluid through an organ or tissue while it remains in its anatomical location, enabling targeted support, assessment, or treatment. Clinicians typically access the organ’s supplying vessels and deliver perfusate under regulated pressure, temperature, and flow; the fluid can provide oxygen and nutrients while removing metabolic waste or carrying therapeutic agents. In clinical research and practice, in situ perfusion helps preserve organ viability, evaluate tissue function, and study regional responses to drugs or injury. By maintaining native vascular connections, it can provide physiologically relevant information that isolated or static preparations may not capture.

Ex Situ Normothermic Machine Perfusion of Donor Livers

Here we present a protocol describing oxygenated ex situ machine perfusion of donor liver grafts. This article contains a step by step protocol to procure and prepare the liver graft for machine perfusion, prepare the perfusion fluid, prime the perfusion machine and perform oxygenated normothermic machine perfusion of the liver graft.

Isolation of Rat Portal Fibroblasts by In situ Liver Perfusion

A technique for isolating portal fibroblasts from rat liver is described. Livers are perfused and digested in situ with collagenase, followed by ex vivo digestion of the liver slurry and size selection of cells. This method provides a pure population of portal fibroblasts without the need for passage in culture.
