Macrophage Recruitment

Macrophage recruitment is the directed movement of macrophages or their circulating precursors toward damaged, infected, or diseased tissue, where they shape local immune responses. Inflammatory signals such as chemokines and cytokines create concentration gradients that guide chemotaxis, while adhesion molecules help cells attach to and cross the vascular endothelium before entering tissue. This process supports pathogen clearance, debris removal, tissue repair, and resolution of inflammation, but excessive or prolonged recruitment can contribute to chronic inflammatory disease and tumor progression. Understanding its regulation helps researchers identify biomarkers and develop therapies that enhance repair or limit harmful immune activity.

The macrophages are first primed with bacterial lipopolysaccharide (LPS) to upregulate NLRP3 protein expression. Exposing the macrophages to nigericin—a potassium ionophore—leads to the activation of NLRP3 and the formation of inflammasomes, which are visualized via immunofluorescence.

The article describes a readily easy adaptive in vitro model to investigate macrophage polarization. In the presence of GM-CSF/M-CSF, hematopoietic stem/progenitor cells from the bone marrow are directed into monocytic differentiation, followed by M1 or M2 stimulation. The activation status can be tracked by changes in cell surface antigens, gene expression and cell signaling pathways.
