Blood Cell Differentiation

Blood cell differentiation is the developmental process by which hematopoietic stem cells generate the diverse cell types found in blood, including erythrocytes, leukocytes, and platelets. It begins when stem cells self-renew or commit to myeloid or lymphoid lineages in response to signals from the bone marrow microenvironment, cytokines, and lineage-specific transcription factors that regulate gene expression. Studying this process in developmental biology reveals how blood and immune systems form, mature, and maintain balanced cell populations. It also supports research into leukemia, anemia, immune disorders, stem cell transplantation, and regenerative medicine by identifying how abnormal or incomplete differentiation disrupts blood function.

In this video, we demonstrate a procedure to induce the differentiation and polarization of peripheral blood-derived monocytes into M1 or M2 macrophages. Incubation with granulocyte-macrophage colony-stimulating factor, lipopolysaccharide, and interferon‐gamma yields pro-inflammatory M1 macrophages, while incubation with macrophage colony-stimulating factor and interleukin-4 induces anti-inflammatory M2 macrophage production.
