Cell Persistence Biodistribution

Cell persistence and biodistribution describe how long introduced cells survive and where they localize, migrate, or become retained within an organism, making them essential for understanding cell fate in developmental biology. These patterns arise from interacting processes such as tissue homing, engraftment, proliferation, apoptosis, immune-mediated clearance, and signals from the local microenvironment. Researchers assess persistence and distribution with cell labeling, imaging, and tissue-based analyses to determine whether cells reach intended developmental sites and maintain their identity. Such measurements clarify lineage behavior, tissue integration, and the performance and safety of cell-based models or regenerative strategies.

2023

Source: Peiman Shahbeigi-Roodposhti and Sina Shahbazmohamadi, Biomedical Engineering Department, University of Connecticut, Storrs, Connecticut Nanoparticles have been increasingly used research towards targeted drug delivery and controlled drug release. While most of these particles have been developed as polymeric or liposomal particles because of their biocompatibility, there is a trend in current research toward the use of metallic and magnetic nanoparticles. These metallic nanoparticles...

In Vitro Persister Assay: A Method to Evaluate the Effect of Osmolytes on Bacterial Persistence

2025

In this video, we demonstrate a method to evaluate the effect of osmolytes on the formation of bacterial persisters. The bacterial cells are first exposed to osmolytes to induce osmotic stress, triggering persister formation, then treated with antibiotics to enumerate the persisters.

Assessing Cell Layer Permeability in Non-Replicating Persistent Mycobacterium tuberculosis

2026

Begin with tubes containing the non-replicating persistent, or NRP stage of Mycobacterium tuberculosis, which has a thickened outer layer.Add glass beads to one tube and shake it to mechanically disrupt the mycobacteria’s outer layer, enhancing permeability. Next, transfer the bead-beaten mycobacterial suspension to a fresh tube.Add fluorescently-labeled rifampicin, an anti-tuberculosis antibiotic, to both the untreated and bead-beaten cell suspensions.Incubate with shaking to allow...

An In Vivo Immunofluorescence Localization Method to Study Antibody Biodistribution

2025

This video demonstrates an in vivo immunofluorescence localization (IVIL) technique to study the biodistribution of tumor-targeting antibodies in a murine model of breast cancer. Upon the intravenous injection of a tumor-specific antibody in the mouse, the tumor is surgically isolated, and the localization of the antibody within the tumor tissue is confirmed via ex vivo immunostaining.

Acute and Chronic Models of Hyperglycemia in Zebrafish: A Method to Assess the Impact of Hyperglycemia on Neurogenesis and the Biodistribution of Radiolabeled Molecules

2017

This work describes methods to establish acute and chronic hyperglycemia models in zebrafish. The aim is to investigate the impact of hyperglycemia on physiological processes, such as constitutive and injury-induced neurogenesis. The work also highlights the use of zebrafish to follow radiolabeled molecules (here, [18F]-FDG) using PET/CT.
