Gland Formation Ability

Gland formation ability is the capacity of developing cells or tissues to organize into specialized secretory organs, a key feature of organogenesis in developmental biology. It typically depends on coordinated epithelial differentiation, tissue invagination or budding, and reciprocal signaling between epithelial and surrounding mesenchymal cells that guides branching, lumen formation, and functional maturation. Studying this ability helps explain how endocrine and exocrine glands arise, how developmental signals shape organ architecture, and how disruptions can contribute to congenital abnormalities. These insights also support research on tissue regeneration, disease modeling, and organoid systems that recreate gland development in vitro.

In this video, we perform a clonogenic assay of keratinocytes harvested from adult mice to determine the ability of individual cells to proliferate and form colonies. This keratinocyte harvesting technique is useful for studying other downstream aspects of cutaneous chemical carcinogenesis and tumor promotion.

This protocol describes methods for purifying, quantitating, and characterizing extracellular vesicles (EVs)/exosomes from non-adherent/mesenchymal mammary epithelial cells and for using them to transfer mammary gland-forming ability to luminal mammary epithelial cells. EVs/exosomes derived from stem-like mammary epithelial cells can transfer this cell property to cells that ingest the EVs/exosomes.
