Hand Stripping

Hand stripping is a method for manually collecting mature gametes from reproductive animals, especially fish, by applying controlled pressure to release eggs or sperm. In developmental biology, researchers gently massage the abdomen of a ripe female or male so gametes are expressed into separate, clean containers, then combine them under controlled conditions for external fertilization. This approach provides synchronized access to early embryos and supports studies of fertilization, cleavage, gastrulation, organ formation, and developmental abnormalities. It is also useful for hatchery breeding, maintaining genetic lines, and producing experimental embryos for developmental, genetic, and toxicological research.

This protocol describes the development of two IgG class monoclonal antibodies (mAbs) strongly reactive to myoglobin of cetaceans. These mAbs are applied on a colloidal gold immunochromatographic test strip based on the sandwich format to differentiate the Mb of cetaceans from seal and other animals.
