Metabolic Respiration Analysis

Metabolic respiration analysis is the quantitative assessment of how cells produce energy through pathways that consume oxygen or release carbon dioxide, providing a window into cellular function. In practice, researchers measure oxygen consumption and related changes in extracellular acidity to distinguish mitochondrial oxidative phosphorylation from glycolytic activity under defined developmental or environmental conditions. These measurements reveal how energy production shifts as cells proliferate, differentiate, migrate, or respond to stress. In developmental biology, the approach helps link metabolic state with tissue formation, cell fate decisions, and disease-associated developmental defects.

Zebrafish represent a powerful vertebrate model that has been under-utilised for metabolic studies. Here we describe a rapid way to measure the in vivo metabolic profile of developing zebrafish that allows the comparison of different mitochondrial function parameters between genetically or pharmacologically manipulated embryos, thereby increasing the applicability of this organism.

Quantifying Respiration using Microrespirometers ExpandNOTE: In this experiment, you will measure the rate of cellular respiration for germinating seeds by measuring the rate of exchange for oxygen. As oxygen is consumed to provide energy, germinating seeds release carbon dioxide. This carbon dioxide is absorbed by potassium carbonate and thus the overall gaseous pressure of the respirometer will be reduced. HYPOTHESES: The experimental hypothesis is that germinating seeds will show a greater...

Autotrophs and Heterotrophs Living organisms require a continuous input of energy to maintain cellular and organismal functions such as growth, repair, movement, defense, and reproduction. Cells can only use chemical energy to fuel their functions, therefore they need to harvest energy from chemical bonds of biomolecules, such as sugars and lipids. Autotrophic organisms, namely plants, algae, and photosynthetic and chemosynthetic bacteria, convert inorganic materials into such biomolecules by...

Begin with a slurry containing bacterial cells treated with silver nanoparticles suspended in an exopolysaccharide matrix.Add a phosphate buffer to the slurry to maintain a stable pH.Next, introduce a solution containing glucose and a soluble redox indicator, then incubate the mixture. The redox indicator acts as an electron acceptor.During incubation, glucose and the redox indicator enter metabolically active bacterial cells. The cells utilize glucose during respiration, generating...
