Polymer Infusion

Polymer infusion is a technique that introduces a liquid polymer into biological tissues, cavities, or microvascular networks so their three-dimensional structure can be preserved for analysis. A low-viscosity polymer penetrates accessible spaces and then cures or polymerizes, forming a stable cast or infiltrated matrix that retains the specimen’s internal architecture. In developmental biology, this approach can help visualize changing vessel patterns, tissue organization, and anatomical connections in developing embryos or organs. Polymer-infused specimens support microscopy, structural reconstruction, and comparisons of growth-related changes, providing a durable representation of complex biological spaces that may be difficult to observe directly.

Here, we describe protocols for the preparation of trans-cyclobutane fused cyclooctenes (tCBCO), their polymerization to prepare depolymerizable olefinic polymers, and the depolymerization of these polymers under mild conditions. Additionally, protocols for the preparation of depolymerizable networks and compression molding of rigid linear plastics based on this system are described.
