Induced Cardiomyocyte-like Cells

Induced cardiomyocyte-like cells are cells generated by reprogramming noncardiac cells toward a cardiac muscle-like identity, providing a model for studying cell fate during heart development. This process typically uses cardiac transcription factors or related molecular signals to alter gene expression, activate cardiomyocyte-associated programs, and induce features such as sarcomeric organization and contractile activity. In developmental biology, these cells help reveal how cellular identity is established and maintained, while offering potential applications in cardiac disease modeling, drug testing, and regenerative research. Their degree of maturation and functional similarity to native cardiomyocytes remains an important focus for improving their scientific and therapeutic value.

Pluripotent stem cells, either embryonic or induced pluripotent stem (iPS) cells, constitute a valuable source of human differentiated cells, including cardiomyocytes. Here, we will focus on cardiac induction of iPS cells, showing how to use them to obtain functional human cardiomyocytes through an embryoid bodies-based protocol.

Here, we describe a robust protocol for human cardiomyocyte derivation that combines small molecule-modulated cardiac differentiation and glucose deprivation-mediated cardiomyocyte purification, enabling production of purified cardiomyocytes for the purposes of cardiovascular disease modeling and drug screening.

We describe here a protocol for the generation of iCMs using retrovirus-mediated delivery of Gata4, Tbx5 and Mef2c in a polycistronic construct. This protocol yields a relatively homogeneous population of reprogrammed cells with improved efficiency and quality and is valuable for future studies of iCM reprogramming.

To distinguish cell division from cell cycle variations in cardiomyocytes, we present protocols using two transgenic mouse lines: Myh6-H2B-mCh transgenic mice, for the unequivocal identification of cardiomyocyte nuclei, and CAG-eGFP-anillin mice, for distinguishing cell division from cell cycle variations.
