Ureteric Bud Branching

Ureteric bud branching is the developmental process through which an epithelial outgrowth from the Wolffian duct repeatedly divides to form the collecting duct system of the kidney. It begins when signals from the metanephric mesenchyme, particularly glial cell line-derived neurotrophic factor (GDNF) acting through RET receptors, stimulate bud outgrowth, while reciprocal epithelial-mesenchymal signaling guides branch formation and nephron induction. This branching morphogenesis establishes the kidney’s collecting network and coordinates nephron development during embryogenesis. Studying the process helps explain congenital anomalies of the kidney and urinary tract and supports research using kidney organoids and regenerative models.

Glycogen Branching Assay to Measure the Degree of Glycogen Branching

In this video, we describe a qualitative spectrophotometric assay to determine the extent of branching in a glycogen sample with an uncharacterized structure. Comparing the absorbance maxima of the uncharacterized sample with that of polysaccharides with known branched structures indicates the presence of branching in the uncharacterized sample.

The murine model of irreversible unilateral ureteric obstruction (UUO) is presented together with the model of reversible UUO in which the ureteric obstruction is relieved by anastomosis of the severed ureter into the bladder. These models enable the study of renal inflammation and scarring as well as tissue remodeling.

The surface of the tongue is covered with various small bumps called papillae, which either distribute what has been ingested (filiform papillae) or contain the sensory taste (or gustatory) receptor cells (fungiform, circumvallate, and foliate papillae). Embedded within each taste-related papilla are the taste buds—clusters of 30 to 100 gustatory receptor cells. Gustatory receptor cells extend finger-like projections called gustatory hairs (or microvilli) into a region known as the taste pore.
