Cyclic Wet Dry

Cyclic wet-dry exposure is an engineering test condition in which a material or structure undergoes repeated periods of moisture and drying to evaluate durability under changing environmental conditions. During each cycle, water enters pores, joints, or surface defects, while drying drives evaporation and often produces dimensional changes, chemical concentration, or stress gradients that can initiate cracking, corrosion, or surface deterioration. Engineers apply these cycles to concrete, soils, coatings, metals, and other construction materials to simulate service environments, compare protective treatments, and identify failure mechanisms. Results support material selection, service-life prediction, and the design of more resilient infrastructure.

We describe a framework incorporating straightforward biochemical and computational analysis to guide the characterization and crystallization of large coiled-coil domains. This framework can be adapted for globular proteins or extended to incorporate a variety of high-throughput techniques.
