Load Fluctuation

Load fluctuation is the time-dependent variation in demand placed on an engineering system, such as changes in electrical power consumption, mechanical force, or process throughput. As the load rises or falls, the system’s components must respond to altered energy, torque, pressure, or current requirements; rapid changes can produce transient behavior, stress, vibration, or instability when control and storage capacity are insufficient. Engineers analyze load profiles and fluctuations to size equipment, design feedback controls, maintain voltage or speed, reduce fatigue, and improve reliability. This concept is central to power grids, rotating machinery, structural design, transportation systems, and industrial automation.

This video demonstrates the technique of fluorescence fluctuation spectroscopy (FFS) to study protein homo-oligomerization. Fluorescently-labeled proteins in a sample are dimerized using an agent when studying protein oligomerization using FFS. Using a confocal microscope, as the proteins move in and out of the small observation volume, the analysis of the fluctuations in the brightness of the fluorescent molecules is performed to determine the oligomeric state of the proteins.
