Environmental Water Samples

Environmental water samples are collected portions of natural or human-impacted waters used to assess water quality and environmental conditions. Sampling plans target locations, depths, and times that represent rivers, lakes, groundwater, or wastewater, while appropriate containers, preservation, and transport help maintain the chemical and biological properties of each sample. Laboratory analysis can measure physical characteristics, nutrients, contaminants, and microorganisms, linking observed results to pollution sources and ecosystem change. These samples support environmental monitoring, regulatory assessment, pollution control, public health protection, and research on how human activities affect aquatic systems.

We describe a protocol for filtration of water samples with a filter cartridge and extraction of environmental DNA (eDNA) without having to cut open the housing to remove the filter. This protocol is developed for metabarcoding eDNA from fishes, but is also applicable to eDNA from other organisms.

This manuscript describes an automated gel size selection approach for purifying DNA fragments for next-generation sequencing. The Ranger Technology provides complete automation of the entire process of agarose gel loading, electrophoretic analysis, and recovery of targeted DNA fragments allowing for high-throughput and high quality next-generation sequencing libraries.
