Its Region Amplification

ITS region amplification is a molecular technique used to copy the internal transcribed spacer (ITS) region of ribosomal DNA, a variable genetic marker commonly used to identify fungi and other environmental microorganisms. In polymerase chain reaction (PCR), primers bind to conserved sequences flanking the ITS region, and repeated cycles of denaturation, primer annealing, and DNA extension generate many copies for analysis. Researchers can then sequence or compare the amplified products to characterize fungal communities in soil, water, plant-associated environments, and other samples. The method supports biodiversity surveys, ecological monitoring, and detection of environmental changes affecting microbial communities.

Telomerase is expressed in the neonatal brain and also in distinct regions of the adult brain. We present a non-toxic time saving TRAP assay for the analysis of telomerase activity in various regions of the mouse brain and detection of differences in telomerase activity between male and female mouse brains.

Source: Tomás, J. M., et al. Generation of Null Mutants to Elucidate the Role of Bacterial Glycosyltransferases in Bacterial Motility. J. Vis. Exp. (2022).This video demonstrates the initiator-dependent amplification of a suicide plasmid in E. coli. The plasmid, which carries an antibiotic resistance gene, replicates only in the presence of an initiator protein encoded by the host. After electroporation and incubation in recovery medium, the mixture is plated on antibiotic-containing agar.
