Minimal Media Agar

Minimal Media Agar is a solid growth medium containing only the essential nutrients required by microorganisms, making it useful for studying their nutritional needs and metabolic capabilities. Agar provides a stable surface for colony formation, while the chemically defined nutrient composition limits growth to organisms able to synthesize missing compounds or use the available carbon and energy sources. In environmental microbiology, this medium helps isolate and characterize microorganisms from soil, water, and other samples, including organisms adapted to nutrient-poor conditions. Growth patterns can reveal metabolic diversity, support strain selection, and inform studies of microbial ecology and biogeochemical processes.

Agar-Block Microcosms for Controlled Plant Tissue Decomposition by Aerobic Fungi

2011

This video demonstrates a controlled environment approach to study degradation of lignocellulosic plant tissues by aerobic fungi. The ability to control nutrient sources and moisture is a key advantage of agar-block microcosms, but the approach often yields mixed success. We address critical pitfalls to yield reproducible, low-variability results.

Assay for Adhesion and Agar Invasion in S. cerevisiae

We describe a qualitative assay for yeast adhesion and agar invasion as a measure of invasive and pseudohyphal differentiation. This simple assay can be used to assess the invasive phenotype of various mutants as well as the effects environmental cues and signaling pathways on yeast differentiation.

A Charcoal Agar Resazurin Assay to Evaluate Test Compound Activity Against Mycobacteria

2025

This video showcases the Charcoal Agar Resazurin Assay (CARA) for evaluating the activity of antimicrobial compounds against Mycobacterium tuberculosis. CARA aids in assessing the efficacy of these compounds by gauging their impact on bacterial growth and distinguishing between bactericidal and bacteriostatic effects.

Subculturing Sulfite-Reducing Anaerobic Bacteria from an Agar Culture

Source: Anwar, Z., et al. Enrichment and Detection of Clostridium perfringens Toxinotypes in Retail Food Samples. J. Vis. Exp. (2019).In this video, sulfite-reducing bacteria are cultured in a sulfite-containing agar medium, where they grow anaerobically and reduce sulfite to hydrogen sulfide, forming black precipitates with iron ions. Black colonies are isolated from the agar depth using aseptic techniques and transferred to fresh medium for continued anaerobic incubation.

Development of a More Sensitive and Specific Chromogenic Agar Medium for the Detection of Vibrio parahaemolyticus and Other Vibrio Species

Detection and isolation of clinically relevant Vibrio species require selective and differential culture media. This study evaluated the ability of a new chromogenic medium to detect and identify V. parahaemolyticus and other related species. The new medium was found to have better sensitivity and specificity than the conventional medium.
