Differential Labeling

Differential labeling is a comparative method that assigns distinct, measurable labels to samples, organisms, or environmental materials so their identities, origins, or fates can be distinguished. Labels may produce different chemical, isotopic, fluorescent, or other analytical signals, allowing researchers to track labeled components against an unlabeled or differently labeled reference under controlled conditions. In environmental research, this approach helps trace nutrient movement, identify contaminant sources, follow microbial or plant responses, and separate overlapping pathways in complex ecosystems. By linking a detectable signal to a specific source or treatment, differential labeling strengthens measurements of transport, transformation, uptake, and environmental change.

Reconstitution of Nucleosomes with Differentially Isotope-labeled Sister Histones

This protocol describes the reconstitution of nucleosomes containing differentially isotope-labeled sister histones. At the same time, asymmetrically post-translationally modified nucleosomes can be generated after using a premodified histone copy. These preparations can be readily used to study modification crosstalk mechanisms, simultaneously on both sister histones, by using high-resolution NMR spectroscopy.

Differential Labeling of Cell-surface and Internalized Proteins after Antibody Feeding of Live Cultured Neurons

We describe a method to label protein on the surface of living neurons using a specific polyclonal antibody to extracellular epitopes. Protein bound by the antibody on the cell surface and subsequently internalized via endocytosis can be distinguished from protein remaining on, or trafficked to, the surface during the incubation.
