Dna Quality Assessment

DNA quality assessment is the evaluation of nucleic acid purity, concentration, integrity, and suitability for downstream analysis. It combines absorbance measurements, fluorescence-based quantification, gel electrophoresis, and amplification tests to identify degradation, contamination, or PCR inhibitors in extracts from soil, water, sediments, and organisms. In environmental sciences, reliable assessment helps distinguish poor sampling or extraction from genuine biological patterns in metabarcoding, environmental DNA surveys, and microbial community studies. Consistent quality-control measurements improve reproducibility, reduce false negatives and biased profiles, and support confident interpretation of biodiversity, pollution, and ecosystem-monitoring results.

We present a noninvasive sampling approach to efficiently collect hair samples from elusive small mammals, as shown for the American pika. We demonstrate the utility of this method by extracting DNA from sampled hair and amplifying several types of molecular markers commonly used in studies of wildlife ecology and conservation.

Here, we present a protocol for tracing genomic DNA (gDNA) contamination in RNA samples. The presented method utilizes primers specific for the internal transcribed spacer region (ITS) of ribosomal DNA (rDNA) genes. The method is suited for reliable and sensitive detection of DNA contamination in most eukaryotes and prokaryotes.

The combination of chromatin immunoprecipitation and ultra-high-throughput sequencing (ChIP-seq) can identify and map protein-DNA interactions in a given tissue or cell line. Outlined is how to generate a high quality ChIP template for subsequent sequencing, using experience with the transcription factor TCF7L2 as an example.
