Allelic Exchange

Allelic exchange is a genetic engineering method that replaces a specific chromosomal DNA sequence with a designed alternative, enabling precise analysis of gene function. It typically uses a donor DNA construct containing regions homologous to the target locus, allowing homologous recombination to integrate the altered allele; selectable markers and, in some systems, counterselection help identify cells carrying the intended replacement. In genetics, this approach supports targeted gene deletions, insertions, point mutations, and sequence corrections, particularly in bacteria and other organisms amenable to homologous recombination. By linking defined DNA changes to observable phenotypes, allelic exchange advances functional genomics, microbial physiology, and studies of genetic regulation.

Multiple Allele Traits

For the same gene multiple alleles can interact to influence phenotypes like the shape and protein composition of an individual cells.By studying allele interactions on the molecular and cellular levels researchers can understand the resulting phenotypes and complications of human conditions like sickle cell trait, improving treatment. The ABO blood group system is a common example of multiple alleles in humans. This system includes three alleles called IA, IB, and i alleles, which combine in...

Agouti: A Lethal Allele Lucien Cuénot discovered lethal alleles in 1905 while studying the inheritance of coat color in mice. The agouti gene is responsible for the color of the coat in mice. This gene codes for an agouti-signaling protein, which is responsible for melanin distribution in mammals. The wild-type allele gives rise to gray-brown coat color in mice, while the mutant allele gives rise to yellow coat color. In addition to coat color, the agouti gene is associated with the yellow...

Understanding the Development of Compensatory Pathways in a Mutant Malaria Parasite Harbouring Hypomorphic Allele of Plant-Like Kinases

Chemical genetics involves the substitution of a gatekeeper residue with an amino acid containing a different side chain at the target locus. Here, we have generated a mutant parasite containing a hypomorphic allele of cdpk1 and identified compensatory pathways adopted by the parasite in the mutant background.
