Bidirectional Pcr Methodology

Bidirectional PCR methodology is a polymerase chain reaction approach that amplifies a defined DNA region from both directions, enabling targeted analysis of sequences in genetics. It uses two primers that bind to opposite DNA strands and repeated cycles of denaturation, primer annealing, and DNA polymerase-mediated extension to produce exponential amplification of the intervening fragment. The resulting amplicon can support genotyping, mutation detection, sequence verification, and analysis of genetic variation. By generating substantial quantities of a specific DNA target from limited starting material, bidirectional PCR provides a sensitive and adaptable foundation for molecular diagnostics, genetic research, and downstream sequencing workflows.

This manuscript describes the experimental procedure and software analysis for a bidirectional integration site assay that can simultaneously analyze upstream and downstream vector-host junction DNA. Bidirectional PCR products can be used for any downstream sequencing platform. The resulting data are useful for a high-throughput, quantitative comparison of integrated DNA targets.
