Plant Microrna Annotation

Plant microRNA annotation is the process of identifying and characterizing genomic sequences that produce microRNAs, small regulatory RNAs that influence gene expression and development. It combines genome analysis, small-RNA sequencing, precursor prediction, and secondary-structure assessment to distinguish authentic microRNA loci from other short RNA sequences, often using conserved sequence and expression evidence. In genetics, annotation helps link microRNAs with their predicted messenger RNA targets and regulatory pathways. These data support studies of plant growth, stress responses, development, and heritable variation, while improving the interpretation of plant genomes and guiding research into crop traits and gene-regulatory networks.

2026

Source: Zhao, J., et al. Potato Virus X-Based microRNA Silencing (VbMS) In Potato. J. Vis. Exp. (2020).This video demonstrates the use of Agrobacterium tumefaciens to deliver a Potato Virus X (PVX)-based construct into in vitro-grown potato plants for virus-based microRNA silencing (VbMS). The method enables systemic suppression of a target microRNA, allowing researchers to study its role in plant development through observable phenotypic changes.

MicroRNAs

MicroRNA (miRNA) are short, regulatory RNA transcribed from introns—non-coding regions of a gene—or intergenic regions—stretches of DNA present between genes. Several processing steps are required to form biologically active, mature miRNA. The initial transcript, called primary miRNA (pri-mRNA), base-pairs with itself forming a stem-loop structure. Within the nucleus, an endonuclease enzyme, called Drosha, shortens the stem-loop structure into hairpin-shaped pre-miRNA. After the pre-miRNA ends...
