Red System Proteins

Red system proteins are bacteriophage λ recombination factors that promote targeted DNA exchange in bacterial cells, making them important tools in genetic engineering. The system typically combines Gam, which inhibits the host RecBCD nuclease, with Exo, a 5′-to-3′ double-stranded DNA exonuclease, and Beta, which binds the resulting single-stranded DNA and promotes annealing to a homologous genomic sequence. Together, these proteins support homologous recombination without requiring extensive selectable markers or long cloning steps. In genetics, Red system proteins enable recombineering, allowing researchers to introduce precise mutations, insertions, deletions, and sequence replacements for functional studies and genome construction.
