Sybr Green Detection

SYBR Green detection is a fluorescence-based method for measuring double-stranded DNA, widely used in genetic analysis and real-time PCR. The dye binds to double-stranded DNA and produces a much stronger fluorescent signal than in its free form; during PCR, fluorescence increases as newly synthesized DNA accumulates, allowing amplification to be monitored cycle by cycle. Researchers use the resulting signal to estimate starting template abundance and assess gene expression after reverse transcription, while melting-curve analysis can help distinguish the intended amplicon from primer-dimers or other nonspecific products. Because the dye binds any double-stranded DNA, careful primer design and controls are essential.

This protocol diagnoses Tilapia Lake Virus (TiLV) in tilapia tissues using RT-PCR methodologies. The entire method is described from tissue dissection to total RNA extraction, followed by cDNA synthesis and detection of TiLV by either conventional PCR or quantitative PCR using dsDNA binding a fluorescent binding dye.

We describe a molecular clone of maedi-visna virus that expresses GFP and is fully infectious. Replication of this virus can be detected by using fluorescence microscopy and flow cytometry.
