Antibody Phagocytosis

Antibody phagocytosis is an immune process in which antibodies mark microbes or other targets for engulfment and destruction by phagocytic cells, such as macrophages and neutrophils. Antibodies bind antigens on a target’s surface, while their exposed Fc regions engage Fc receptors on phagocytes, triggering attachment, membrane extension, and formation of an intracellular phagosome that can fuse with lysosomes for degradation. This opsonization mechanism helps control bacterial and fungal infections and links adaptive antibody responses with innate cellular defenses. Studying antibody phagocytosis clarifies immune protection, informs vaccine assessment, and supports research into antibody-based therapies and immune evasion.

An In Vitro Assay to Measure Antibody-Dependent Cellular Phagocytosis by Phagocytes

2025

This video demonstrates an assay to measure the antibody-dependent cellular phagocytosis (ADCP) by breast milk phagocytes. Fluorescently labeled microspheres conjugated to an antigen-antibody complex, are combined with a cell suspension isolated from breast milk to induce antibody-dependent cellular phagocytosis by the phagocytic leukocytes. Upon labeling the cells with antibodies against common leukocyte antigen (CD45), flow cytometry is used to measure the fluorescence intensity of...
