Viral Titration Assay

A viral titration assay is a laboratory method for measuring the concentration of infectious virus in a sample, a key step in characterizing viral infectivity and evaluating immune responses. The assay typically involves serially diluting a virus preparation, exposing susceptible cells to each dilution, and quantifying infection through plaque formation, cytopathic effects, or another detectable signal; results may be reported as plaque-forming units or a 50% tissue culture infectious dose. In immunology and infection research, viral titration supports vaccine and antiviral studies, defines experimental doses, and enables comparisons of viral replication across samples and conditions.

Source: Bouillier, C., et al. Generation, Amplification, and Titration of Recombinant Respiratory Syncytial Viruses. J. Vis. Exp. (2019)The video demonstrates a plaque titration assay used to quantify infectious recombinant respiratory syncytial virus (RSV) in epithelial cell monolayers. It presents the infection of host cells with serially diluted viral suspensions, the restriction of viral spread by a cellulose-based semi-solid overlay, and the formation of localized plaques from cytopathic...

2023

Source: Laboratory of Dr. Yee Nee Tan — Agency for Science, Technology, and Research Titration is a common technique used to quantitatively determine the unknown concentration of an identified analyte.1-4 It is also called volumetric analysis, as the measurement of volumes is critical in titration. There are many types of titrations based on the types of reactions they exploit. The most common types are acid-base titrations and redox titrations.5-11 In a typical titration process, a standard...
