Bfp To Gfp Conversion

BFP-to-GFP conversion is an optical labeling strategy in which an engineered fluorescent protein changes from blue to green emission, enabling researchers to mark and follow selected cells or microorganisms over time. In photoconvertible systems, controlled short-wavelength illumination induces a structural or chemical change in the chromophore, shifting its fluorescence spectrum without requiring new protein synthesis. This temporal labeling supports fluorescence microscopy and quantitative tracking in immunology and infection research, including studies of immune-cell migration, host-pathogen interactions, microbial spread, and population dynamics. By distinguishing material labeled before conversion from material produced afterward, the method links spatial location with cellular or infectious history.

While the transport of cell surface proteins is relatively easily studied, visualizing the trafficking of intracellular proteins is much more difficult. Here, we use constructs incorporating photoactivatable GFP and demonstrate a method to accurately follow the amyloid precursor protein from the Golgi apparatus to down-stream compartments and follow its clearance.
