Bladder Epithelial Cells

Bladder epithelial cells are urothelial cells that line the inner surface of the urinary bladder, forming a specialized barrier between urine and underlying tissue. Their apical surface contains uroplakins and a glycosaminoglycan layer, while tight junctions limit permeability; pattern-recognition receptors can detect microbial components and trigger cytokine and chemokine release. These properties make bladder epithelial cells central to studying urinary tract infection, including bacterial adherence, invasion, innate immune signaling, and barrier repair, and support evaluation of host-directed therapies and antimicrobial strategies.

Take cultures of human urinary bladder epithelial cells in a multiwell plate.Incubate the control culture with wild-type Enterobacter cloacae, a pathogenic bacterium with pili, hair-like structures on the bacterial surface. The FimH subunit of the pili binds to the host cell receptor, anchoring the wild-type bacteria to the host cell.Incubate the experimental culture with a mutant Enterobacter cloacae lacking FimH.Without FimH, the mutant bacteria fail to adhere to the host...
