Colloidal Gold Preparation

Colloidal gold preparation is the production of stable suspensions of nanoscale gold particles, which provide distinctive optical properties and useful surfaces for biological detection. Typically, chloroauric acid is chemically reduced in solution, while stabilizing agents such as citrate prevent particle aggregation; reagent concentrations, temperature, and mixing conditions influence particle size and dispersion. In immunology and infection research, the resulting particles can be functionalized with antibodies or other ligands to bind specific antigens. These conjugates support immunogold electron microscopy, rapid lateral-flow assays, and other diagnostic formats, enabling visible or high-resolution detection of pathogens, cellular structures, and immune responses.

This protocol describes the development of two IgG class monoclonal antibodies (mAbs) strongly reactive to myoglobin of cetaceans. These mAbs are applied on a colloidal gold immunochromatographic test strip based on the sandwich format to differentiate the Mb of cetaceans from seal and other animals.

Children at play often make suspensions such as mixtures of mud and water, flour and water, or a suspension of solid pigments in water known as tempera paint. These suspensions are heterogeneous mixtures composed of relatively large particles that are visible to the naked eye or can be seen with a magnifying glass. They are cloudy, and the suspended particles settle out after mixing. On the other hand, a solution is a homogeneous mixture in which no settling occurs and in which the dissolved...
