Cytochrome-c Release

Cytochrome-c release is a mitochondrial signaling event in which cytochrome c moves from the intermembrane space into the cytosol, where it helps initiate intrinsic apoptosis and shape immune responses to cellular stress and infection. During mitochondrial outer membrane permeabilization, often controlled by pro- and anti-apoptotic BCL-2 family proteins, released cytochrome c binds APAF1 and ATP or dATP to assemble the apoptosome, activating caspase-9 and downstream executioner caspases. In immunology and infection research, measuring this pathway helps clarify how pathogens, immune signals, and cellular damage influence infected-cell survival, inflammatory outcomes, and host defense.

Activation of Apoptosis by Cytoplasmic Microinjection of Cytochrome c

In this protocol, we describe the direct cytoplasmic microinjection of cytochrome c protein into fibroblasts and primary sympathetic neurons. This technique allows for the introduction of cytochrome c protein into the cytoplasm of cells and mimics the release of cytochrome c from mitochondria, which occurs during apoptosis.

This procedure describes how to encapsulate cytochrome c (cyt. c) in silica (SiO2) sol-gels, process these gels to form bioaerogels, and use these bioaerogels to rapidly recognize nitric oxide (NO) through a gas-phase reaction. This type of protocol may aid in the future development of biosensors or other bioanalytical devices.
