Dissociation Curve Analysis

Dissociation curve analysis is a quantitative method used to characterize how molecular complexes separate under changing conditions, most commonly by measuring the thermal melting of double-stranded DNA after real-time PCR. As temperature rises, complementary DNA strands dissociate, reducing fluorescent dye binding and producing a characteristic melting profile whose peak reflects the amplicon’s melting temperature. In immunology and infection research, this analysis helps assess PCR product specificity, distinguish target sequences from primer-dimers or nonspecific products, and support pathogen detection and genotyping. It provides a rapid quality-control step that strengthens interpretation of nucleic acid amplification results.

Zebrafish are an emerging system for modeling human disorders of the skeletal muscle. We describe a fast and efficient method to isolate skeletal muscle myofibers from embryonic and larval zebrafish.
